I carry the C9 mutation that took my dad’s life with ALS and is coming for mine, and this is the single most promising thing I’m doing about it. A genetic fix for C9, the most common inherited form of ALS, already works in human cells. The barrier now is delivery: we still can’t reliably get gene therapy into the brain and spinal cord, or much of the body beyond the liver, eye, muscle, and blood. That one bottleneck stands between millions of people and treatments that are finally within reach. Dr. Claire Clelland at UCSF built the first CRISPR approach to remove the C9 mutation, the most common genetic cause of ALS and frontotemporal dementia. Through CureC9, a nonprofit program I co-founded under EverythingALS, I’m raising philanthropic support for her lab to crack delivery, working directly in human biology with barcoded gene therapy libraries and the guidance of our Scientific Advisory Board. Solve delivery to the brain, and we open a path to a cure for C9 ALS and FTD, and to therapies for the many diseases held back by the same wall. This is personal. I’m a scientist finishing my MD this year, racing to change this before it reaches me, and it’s why I redirected my whole career to this problem. The work is high-risk and high-reward. It will take years and $12M, but it’s exactly what philanthropy is built to back, alongside funding the lab already has from the NIH. So I’m reaching out.
